زمینه و هدف: محدودیت غذایی و تمرین ورزشی یکی از مداخلات کاربردی برای کاهش وزن و افزایش طول عمر می‌باشد. با این حال، ابهاماتی در رابطه با تأثیر محدودیت غذایی با یا بدون تمرین ورزشی بر توده و قدرت عضلانی وجود دارد و هنوز مسیر پیام‌رسانی و پروتئین‌های درگیر در این زمینه به طور دقیق مشخص نشده‌اند. بنابراین، مطالعه حاضر با هدف تعیین تأثیر محدودیت غذایی با یا بدون تمرین ورزشی بر بیان ژن‌های بتا-کاتنین و گلیکوژن سنتازکیناز۳-بتا در عضله‌ نعلی موش‌های صحرایی نر انجام گردید.
روش‌ کار: ۲۴موش صحرایی نربه شکل تصادفی در سه گروه کنترل، محدودیت غذایی و محدودیت غذایی+تمرین ورزشی جایگزین شدند.طی سه ماه پژوهش،گروه کنترل به صورت آزادانه به آب و غذا دسترسی داشت، در حالی که غذای گروه‌های دارای محدودیت غذایی تا حد۵۰ درصد گروه کنترل محدود شد. گروه محدودیت غذایی+تمرین ورزشی علاوه بر محدودیت غذایی، به مدت ۱۲هفته در برنامه‌ی تمرین ورزشی شرکت کرد. ۴۸ساعت پس از آخرین جلسه تمرین، عضله اسکلتی موش‌های صحرایی استخراج و میزان بیان ژن‌های بتا-کاتنین و گلیکوژن سنتازکیناز۳-بتا با استفاده از روش Real Time-PCR بررسی شد. داده‌های حاصله توسط آزمون تحلیل واریانس یک‌طرفه و تعقیبی توکی تجزیه و تحلیل شدند (۰۵/۰>p).
یافته‌ها: بیان ژن بتا-کاتنین در عضله نعلی گروه محدودیت غذایی و محدودیت غذایی+تمرین ورزشی به طوری غیر معنی‌داری کمتر از گروه کنترل بود(به ترتیب %۳۸ و %۸/۸؛ ۰۵/۰<p). در حالی که بیان ژن گلیکوژن سنتازکیناز۳-بتا در گروه محدودیت غذایی به طور معنی‌داریبیشتر از گروه کنترل و محدودیت غذایی+تمرین ورزشی بود(به ترتیب %۴۴۹ و %۱۷۷؛ ۰۱/۰>p).
نتیجه‌گیری: در کل، ۱۲هفته محدودیت غذایی(۵۰%)، موجب افزایش بیان ژن کلیدی گلیکوژن سنتازکیناز۳-بتا در مسیر پیام‌رسانی Wnt عضله نعلی شد و این ممکن است با کاهش توده و قدرت عضله اسکلتی همراه باشد. با این حال، اضافه شدن تمرین ورزشی به محدودیت غذایی ممکن است این روند را کند نماید.
