<p></p><p> زمینه و هدف: اگر چه بررسیها نشان دادهاند که اندوکانابینوییدها اثراتی مرکزی بر نعوظ دارند، ولی اثر محیطی آنها بر نعوظ نامشخص میباشد. هدف از این مطالعه، بررسی اثر آنانداماید(یک کانابینویید اندوژن) بر پاسخهای شل شدگی القاء شده با تحریک اعصاب غیرآدرنرژیک غیر کولینرژیک(NANC=Non adrenergic Non cholinergic) در بافت کورپوس کاورنوزوم(بافت حیاتی در ایجاد نعوظ) موشهای صحرایی نر بوده است. روش بررسی: کورپوس کاورنوزوم موشها بعد از جدا شدن توسط دایسکت دقیق و گذاشتن در حمام ارگان استاندارد اکسیژنه و حاوی آتروپین(1 میکرومول) و گوانتیدین(5 میکرومول)(به ترتیب جهت بلوک کولینرژیک و آدرنرژیک) و به دنبال ایجاد انقباض توسط 5/7 میکرومول فنیلافرین، توسط تحریک الکتریکی در فرکانسهای 2، 5، 10و 15 هرتز، دچار شل شدگی شدند و نتایج توسط دستگاه الکتروفیزیوگراف ثبت گردید. آنانداماید(1و 3 میکرومول در گروههای مجزا)، 20 دقیقه قبل از تحریکات الکتریکی به حمام ارگان اضافه شد. در گروههای مجزای دیگر، هر یک از آنتاگونیستهای اختصاصی رسپتورهای CB1(یک میکرومول AM251)، CB2(یک میکرومول AM630) و وانیلوییدی(3 میکرومول کاپسازپین)، 45 دقیقه قبل از آنانداماید(یک میکرومول) به حمام اضافه شدند. همچنین، وجود رسپتورهای کانابینوییدی و وانیلوییدی در این بافت توسط روش وسترن بلات ارزیابی شد. هر گروه شامل 6 حیوان بود. نوع مطالعه به صورت تجربی(Experimental) بوده و تحلیل آماری دادهها به وسیله آنالیز یکطرفه واریانس(ANOVA) متعاقب Newman-Keuls as post-hoc test انجام شد و 05/0P< از لحاظ آماری معنیدار در نظر گرفته شد. یافتهها: پاسخهای شلشدگی وابسته به NANC در بافت کورپوس به طور معنیداری(01/0P<) در حضور آنانداماید(1و 3 میکرومول) افزایش یافتند. اثر تقویت کننده آنانداماید بر پاسخهای NANC در حضور AM251(یک میکرومول) و کاپسازپین(3 میکرومول) اما نه با AM630(یک میکرومول)، به طور معنیداری(01/0P<) کاهش یافت. همچنین، مهار کننده آنزیم نیتریک اکساید سنتاز[یک میکرومول L-NAME(Nw-Nitro-L-Arginine Methylester)]، به طور معنیداری(001/0P<) پاســخهای شل شدگــی را در حضــور یا غیــاب آناندامایــد مهار کرد. اگر چه 30 نانومول L-NAME اثری بر پاسخهای NANC نداشت، به طور معنیداری(01/0P<) اثر تقویت کننده آنانداماید را بر پاسخهای NANC کاهش داد. همچنین، آنانداماید اثری بر پاسخهای شل شدگی توسط دوزهای مختلف نیتروپروسایدسدیم(یک میکرومول و 10 نانومول) نداشت. وسترن بلات بافت کورپوس کاورنوزوم نشان داد که پروتئین رسپتورهای CB1 و VR1(اما نه CB2) در این بافت وجود دارند. نتیجهگیری: مطالعه حاضر برای اولین بار نشان داد که آنانداماید(یک اندوکانابینویید) باعث افزایش پاسخهای شلشدگی وابسته به NANC در بافت کورپوس کاورنوزوم موشهای صحرایی نر از طریق هر یک از رسپتورهای CB1 و وانیلوییدی VR1 میشود. به نظر میرسد که در این اثر، مسیر نیتریک اکساید درگیر باشد. همچنین، در این مطالعه نشان داده شد که رسپتورهای CB1 و VR1 در این بافت وجود دارند. </p>
<p></p><p> Background & Aim: Although studies have shown the central effects of Endocannabinoid on erection, its' peripheral effect is unknown. The purpose of this study was to investigate the effect of the endogenous cannabinoid anandamide on the nonadrenergic noncholinergic(NANC) relaxant responses to electrical field stimulation in isolated rat corpus cavernosum, a crucial tissue in erectile function. Material and Methods: The rat corporeal strips were mounted under tension in a standard oxygenated organ bath with guanethidine sulfate(5 µM) and atropine(1 µM)(to produce adrenergic and cholinergic blockade). The strips were precontracted with phenylephrine hydrochloride(7.5 µM) and electrical field stimulation was applied at different frequencies(2, 5, 10, 15 Hz) to obtain NANC-mediated relaxation. Anandamide(0.3, 1 and 3 µM in separate groups) was added 20-min before electrical stimulation. In another group, the selective cannabinoid CB1 receptor antagonist AM251(1 µM), the selective cannabinoid CB2 receptor antagonist AM630(1 µM) and a vanilloid receptor antagonist capsazepine(3 µM) were separately added to the bathing medium 45-min before anandamide(1 µM) administration. Using western blotting, the existence of cannabinoid and vanilloid receptors were assessed in this tissue. Each group consisted of six rats. This study was an experimental study. Statistical analysis of the data was performed by one-way analysis of variance(ANOVA) followed by Newman-keuls post hoc test. Statistical significance was considered when P<0.05. Results: The results showed that the NANC relaxant responses were significantly enhanced in the presence of anandamide at 1 and 3 µM. The potentiating effect of anandamide(1 µM) on relaxation responses was significantly lessened by either AM251(1 µM) or capsazepine(3 µM), but not by AM630(1 µM)(P<0.01). Neither of these antagonists had influence on relaxation responses. Preincubation with the nitric oxide synthase inhibitor L-NAME(1 µM) significantly inhibited the relaxation responses in the presence or absence of 1 µM anandamide(P<0.001). Although at 30 nM, L-NAME did not influence NANC responses, it significantly reduced(P<0.01) the attentuating effect of anandamide on NANC responses. Anandamide(1 µM)) had no influence on concentration-dependent relaxant responses to sodium nitroprusside(10nM-1mM), an NO donor. Western blotting revealed the existence of cannabinoid CB1(but not CB2) and vanilloid VR1 receptors in rat corpus cavernosum. Conclusion: For the first time, our results indicated the potentiating activity of anandamide on NANC-mediated relaxation of rat corpus cavernosum through both CB1 and vanilloid receptors. The NO-mediated component of the NANC relaxant responses to electrical stimulation is involved in this enhancement. Also it was shown that CB1 and VR1 receptors are present in this tissue. </p>
