fa بررسی همخوانی سنجش سطح هورمون استرادیول به روش الایزا در دو حالت رقیق شده و بدون رقت در سرم خانم‌های تحت درمان با داروهای محرک تخمک گذاری Evaluation ofcoordination between diluted and undiluted serum estradiol levels using ELISA method inwomenunder treatment with ovulation induction drugs زنان و زایمان Gynecology پژوهشي Research زمینه و هدف: بررسی سطح هورمون استرادیول در سرم خانم های تحت درمان با داروهای محریک تخمک گذاری به منظور اطمینان از وجود میزان مناسب این هورمون جهت دست یافتن به تعداد کافی تخمک در فرآیند درمان ناباروری و همچنین جلوگیری از بروز عوارض سندرم تحریک بیش از حد تخمدان، امری مهم و ضروری می باشد. در موارد بالا بودن هورمون استرادیول، روش های معمول، توانایی اندازه گیری غلظت واقعی مولکول هورمون را ندارند. بررسی میزان حساسیت روشالایزا برای اندازه گیری هورمون استرادیول و تعیین محدوده قابل اطمینان برای این آزمایش، هدف این مطالعه می‌باشد.<o:p /> روش کار: در این مطالعه مقطعی (Cross-Sectional)، تعداد 132 خانم که برای درمان ناباروی به مرکز فوق تخصصی درمان ناباروری ابن سینا مراجعه نموده و با دارو های محرک تخمک گذاری، تحریک شده بودند، مورد بررسی قرار گرفتند. در این مطالعه از کیت استرادیول الایزا DRG ساخت کشور آلمان استفاده شد. با توجه به شرایط مراجعینی که مورد مطالعه قرار گرفتند، میزان استرادیول سرم آنها بالاتر از Pg/ml 200 بود. جهت بررسی میزان خطی بودن این روش، نمونه سرم تمام این بیماران با نسبت 1:4 رقیق شد و با استفاده از نرم افزار Excel و رسم نمودار مقایسه شدند.<o:p /> یافته‌ها: مشاهده شد که با افزایش غلظت استرادیول بیش از Pg/ml2000، اختلاف بین نمودار مستقیم و رقیق شده افزایش می یابد و اصطلاحاً خطی بودن نمودار روش بدون ر قت از بین می رود.<o:p /> نتیجه‌گیری: در غلظت‌های استرادیول سرمی بیش از Pg/ml 2000 نیاز به رقیق نمودن نمونه‌ها می باشد و نتایج روش بدون رقت قابل اعتماد نخواهد بود. این موضوع باید در آزمایشگاه های مراکز درمان ناباروری و یا حتی آزمایشگاه های دیگر نیز مورد توجه قرار گیرد که میزان خطی بودن روش های مورد استفاده مشخص گردد تا در هنگام مواجهه با مقادیر بالاتر، رقیق سازی صورت گیرد.<o:p /> Background: Measurement of estradiol level in women under treatment with ovulation induction drugs to make sure about sufficient ovum number in the infertility treatment protocol and also to prevent OHSS phenomenon, is an important and necessary factor. In case of high levels of Estradiol hormone, routine methods are unable to detect the real concentration of the hormone molecules. We are aimed to demonstrate the sensitivity of the ELISA method for Estradiol measurement and reliable range of this test.<o:p /> Methods: In this cross-sectional study, 132 women were studied who have appealed to Avicenna infertility clinic for infertility treatment and have stimulated with ovulation induction drugs. DRG ELISA Estradiol kit made in Germany was used in this study. According to the studied population, serum Estradiol levels were higher than 200Pg/ml. To check the linearity of this method, serum samples of all these patients diluted 1:4 and were compared using EXCEL software and charts. <o:p /> Results: It was observed that, in Estradiol hormone concentrations higher than 2000 Pg/ml, the difference between the diluted and undiluted graphs increased and the linearity of the graph disappeared without dilution.<o:p /> Conclusions: Dilution is necessary for the serum Estradiol concentrations more than 2000 Pg/ml and the results without dilution is not reliable. It should be considered in all infertility laboratories and also in other labs that the test linearity should be appointed in each lab to be applied in cases with higher concentrations. <o:p /> استرادیول، ELISA، ناباروری، تحریک تخمک‌گذاری، ایران.
