<p style="DIRECTION: rtl" dir="rtl">نیتریک اکساید( NO ) مولکول کوچک تقریباً ناپایدار و رادیکال آزاد چربی دوستی است که توسط سلولهای متفاوت پستانداران ساخته میشود و در بدن آنها دارای اعمال متفاوتی از جمله واسطه عصبی و اثر سیتوتوکسیک میباشد. این مولکول در مجرای معدی رودهای به عنوان واسطه شیمیایی اعصاب غیر آدرنرژیکی غیر کولینرژیکی( NANC ) عمل میکند و این اعصاب مهاری مسئول شل شدن عضله صاف در این سیستم هستند. علاوه بر آن NO روی سلولهای عضله صاف از طریق اثر در فاز G1 سیکل سلولی اثر مستقیم دارد. این احتمال وجود دارد که در تنگی هیپرتروفیک پیلور در کودکان( IHPS ) که یکی از اختلالات شایع دوران نوزادی میباشد، کمبود ساختــه شدن NO یکــی از علل ایجاد کننده آن باشـــد. در ایــن مطالعـه جهت بررسی اثر نیتریک اکساید روی لایه عضلانی حلقوی پیلور جنین موش صحرایی از ماده مهار کننده تولید NO ( N-nitro-L-arginine methyl ester=L-NAME ) استفاده شد. به این ترتیب که L-NAME بطور روزانه در نرمال سالین حل شده و به موشهای صحرایی باردار نژاد Sprague-Dawley در هفته آخر بارداری به میزان 80 و 20 میلیگرم به ازای کیلوگرم به صورت داخل صفاقی تزریق میگردید. سپس جنینها در روز 21 حاملگی از رحم خارج و معده و دوازدهه آنها از بدن درآورده میشد. پس از انجام مراحل ثابت کردن و پاساژ بافتی و تهیه مقاطع سریال 5 میکرونی، رنگآمیزی تریکرومماسون و پاپ نیکولا صورت گرفت و لامها در زیر میکروسکوپ نوری بررسی شدند. تجزیه و تحلیل آماری نتایج حاصل از مطالعات ماکروسکوپی و میکروسکوپی نشان داد که مقدار 80 میلیگرم به ازای کیلوگرم L-NAME ، در گروه تجربی یک، باعث بروز تغییرات معنیداری(01/0 P< ) شامل کاهش رشد داخل رحمی، تخریب اندام تحتانی، هیپرتروفی و هیپرپلازی پیلور میشود. </p><p dir="rtl" >
Nitric oxide (NO) is an unstable small molecule which acts as lipophilic free radicals. It is synthesized in different mammalian cells and thought to be involved in many physiological and pathological conditions. There are many evidences that show NO acts as a neurotransmitter in Non-Adrenergic, Non-Cholinergic (NANC) nerves, innervating smooth muscles of digestive system and also has direct influence on smooth muscle cells. Infantile hypertrophic pyloric stenosis(IHPS) is one of the most common pathological conditions among the neonates. It is thought that any failure in NO synthesis could be one of the reasons of IHPS. Pregnant Sprague-Dawley rats were used in this study. Different doses of the L-NAME (20-80mg/kg) as an NO inhibitor in normal saline was injected intraperitoneal (IP) during the last week of pregnancy period per day. The embryos were removed on the day of expected delivery. The stomach and duodenum were dissected, fixed by Bouin solution and tissue processing was done. By using rotatory microtome, 5μ serial cross sections were obtained and stained with Trichrom-Mason and Pop-Nicola. Statistical analysis of macroscopic and light microscopic findings showed that 80mg/kg of L-NAME causes significant changes in embryos of trial group including: IUGR, hind-limb disruption, embryo absorption, pyloric hypertrophy and hyperplasia. On the basis of these results it is believed that 80mg/kg of L-NAME can be one of the reasons of pyloric stenosis in infants.
