fa بررسی ارتباط پلی‌مورفیسم‌های شایع ژن پذیرنده ویتامین (VDR) D با استعداد ابتلاء به سل ریوی ایمنی‌شناسی Immunology پژوهشي Research <h2 style="MARGIN: 0cm 0cm 0pt DIRECTION: rtl" dir="rtl"><p><span></span></p><p><span>    زمینه و هدف: ابتلاء به بیماری سل علاوه بر مواجهه با مایکوباکتریوم توبرکولوزیس </span><span dir="ltr"></span><span dir="ltr"><span dir="ltr"></span><font face="Times New Roman">(Mycobacterium tuberculosis-MTB)</font></span><span dir="rtl"></span><span lang="AR-SA"><span dir="rtl"></span>،</span><span lang="AR-SA"> تحت تأثیر عوامل محیطی و ژنتیکی میزبان بوده و تنها کمتر از 10% آلوده شدگان علائم بالینی بیماری را بروز می‌دهند. متابولیسم ویتامین </span><font face="Times New Roman"><span dir="ltr">D</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> سبب فعال شدن ماکروفاژها گردیده و از تکثیر درون سلولی مایکوباکتریوم جلوگیری می‌کند. این روند می‌تواند تحت تأثیر پلی‌مورفیسم‌های شایع ژن پذیرنده ویتامین </span><font face="Times New Roman"><span dir="ltr">D</span><span dir="rtl"></span></font><span><span dir="rtl"></span> </span><span dir="ltr"></span><span dir="ltr"><span dir="ltr"></span><font face="Times New Roman">(Vitamin D Receptor –VDR)</font></span><span dir="rtl"></span><span lang="AR-SA"><span dir="rtl"></span> قرار گیرد. در این مطالعه اثر تغییرات ژنتیکی </span><font face="Times New Roman"><span dir="ltr">VDR</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> در ابتلاء به بیماری سل ریوی مورد بررسی قرار گرفته است.</span><h4 style="MARGIN: 0cm 0cm 0pt" dir="rtl"><span>روش بررسی: این مطالعه به صورت </span><font face="Times New Roman"><span dir="ltr">Case-control</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> در جمعیت ایرانی انجام شده است. در این بررسی</span><span lang="FA">، </span><span lang="AR-SA">اثر 4 پلی‌مورفیسم شایع ژن پذیرنده ویتامین </span><font face="Times New Roman"><span dir="ltr">D</span><span dir="rtl"></span></font><span><span dir="rtl"></span> <span lang="AR-SA"><font size="3" face="B Mitra">(این پلی‌مورفیسم‌ها بر مبنای اثر آنزیم‌های محدودگری که آن‌ها را شناسایی می‌کنند، نامگذاری شده‌اند و عبارتند از: </font></span></span><font face="Times New Roman"><span dir="ltr">FokI (F/f)</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span>، </span><font face="Times New Roman"><span dir="ltr">BsmI (B/b)</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span>، </span><font face="Times New Roman"><span dir="ltr">TaqI (T/t)</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> و </span><font face="Times New Roman"><span dir="ltr">ApaI (A/a)</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span>) در 96 بیمار مبتلا به سل ریوی و 122 فرد سالم در گروه کنترل مورد مطالعه قرار گرفت. در این مطالعه از یک تکنیک </span><font face="Times New Roman"><span dir="ltr">PCR-SSP</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> (</span><font face="Times New Roman"><span dir="ltr">Polymerase Chain Reaction with Sequence-Specific Primers</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span>) اصلاح شده توسط آزمایشگاه ما برای تشخیص پلی‌مورفیسم‌های این ژن استفاده گردید. جهت مقایسه بین مبتلایان به سل ریوی و افراد سالم گروه کنترل، از آزمون مجذور کای (</span><font face="Times New Roman"><span dir="ltr">Chi-square</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span>) استفاده شد.<p></p></span></h4><h4 style="MARGIN: 0cm 0cm 0pt" dir="rtl"><span>یافته‌ها: مشخص گردید که فراوانی اللی و ژنوتایپی نواحی پلی مورفیک ژن </span><font face="Times New Roman"><span dir="ltr">VDR</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> در گروه‌های بیماران و افراد سالم اختلاف معنی‌داری با همدیگر ندارند.<p></p></span></h4><h4 style="MARGIN: 0cm 0cm 0pt" dir="rtl"><span>نتیجه‌گیری: این نتایج اهمیت پلی‌مورفیسم‌های ژنتیکی </span><font face="Times New Roman"><span dir="ltr">VDR</span><span dir="rtl"></span></font><span lang="AR-SA"><span dir="rtl"></span> در ابتلاء به بیماری سل را در جمعیت ایرانی غیرمحتمل نشان می‌دهد.<p></p></span></h4></p></h2><p></p> <p></p><p>  <strong>Background and Aim:</strong> In addition to exposure to Mycobacterium tuberculosis (MTB), development of tuberculosis is influenced by environmental and host genetic factors, and clinical disease only occurs in less than 10% of the infected individuals. Vitamin D metabolism leads to activation of macrophages and restricts the intracellular growth of mycobacterium. This effect may be influenced by polymorphisms in the vitamin D receptor (VDR) gene. In the present study we investigated the impact of VDR gene variation in susceptibility to pulmonary tuberculosis. </p><p> <strong> Materials and Methods: </strong>This study was a case-control analysis in an Iranian population. We evaluated four VDR gene polymorphisms [defined by the presence of restriction endonuclease sites for FokI (F/f), BsmI (B/b), TaqI (T/t), and ApaI (A/a)] in 96 patients with tuberculosis and 122 matched healthy controls.
